Second-hand smoke may lead to dementia

Exposure to even second-hand puff can raise a person’s chances of developing dementia, according to researchers.

A new study, by the Cambridge University, has revealed that second-hand smoke can cause damage to the way one’s brain works, making dementia and other neurological conditions, more likely, the ‘British Medical Journal’ reported.

According to the researchers, the connection could be heart disease, a known risk factor for dementia and which can be triggered by smoking.

Lead researcher Dr David Llewellyn said: “Our results suggest that inhaling other people’s smoke may damage brain, impair cognitive functions such as memory, and make dementia more likely.”

In their study, the researchers tested the saliva of 5,000 non-smokers, all above 50 years, for a chemical called cotinine, a by-product of nicotine which can stay in the body for almost two days after inhaling second-hand smoke.

The subjects were then asked to sit a series of tests designed to assess their level of recall and other skills.

And the findings showed those with a high level of the chemical in their bodies were 44 per cent more likely to develop early memory problems, which can be a warning sign for dementia, than those with low levels.

“Given that passive smoking is also linked to other serious health problems like heart disease and stroke, smokers should avoid lighting up near non-smokers. Our findings also support calls to ban smoking in public places,” ‘The Daily Telegraph’ quoted Dr Llewellyn as saying.

NRI scientist using car technology to make synthetic bone

An Indian-origin scientist at the University of Warwick is developing car technology to help produce synthetic bones for the benefit of patients undergoing bone implant surgery.The method is being developed by Dr Kajal Mallick along with postgraduate researcher James Meredith at the university’s Warwick Manufacturing Group.

The researchers believe the method could offer substantial clinical benefits to patients undergoing bone implant surgery.

The technique involves state-of-the-art extrusion of the implant material through a mould, to produce a 3-dimensional honeycomb texture, with uniform pores throughout.

The material can then be sculpted by the surgeon to precisely match the defect. After implantation bone cells will be transported into the implant and begin to form new bone.

“We worked with a Japanese company which manufactures catalytic converters and used their facility to produce samples which we could then test in the laboratory,” explained Mallick.

“We found that we were able to use calcium phosphates – a family of bioceramics that are routinely used in bone implant operations, but by using this technique we were able to improve significantly both the strength and porosity of the implant,” he said.

“At the present time, there is no product available in the market place that satisfies both these key properties simultaneously. It is nearly an ideal scaffold structure for efficient blood flow and formation of new bone cells,” he added.The increased strength of the material means it could be used in spinal surgery, or in revision hip and knee operations, where currently non-degradable materials such as titanium or steel may be used.

The advantage of increased and interconnected porosity is that the implant can quickly be filled with blood vessels, resulting in a more rapid healing process.

“The synthetic bone we are developing is as strong as normal healthy bone yet porous enough to allow bone cells to inhabit it and generate new bone. Over a period of time, we expect the synthetic bone will resorb, leaving only natural bone,” Meredith said:

“I hope that if we can find an industrial partner to take this to market, we will enable treatment of conditions which up to this point have only been possible using metal replacement parts or low strength foam-like bone substitutes,” he said.

Black raspberries can help prevent cancer

Researchers at the Ohio State Comprehensive Cancer Center have revealed that a component of black raspberries can effectively help in cancer prevention.

They found that anthocyanins, a class of flavonoids in black raspberries, inhibited growth and stimulated apoptosis in the esophagus of rats treated with an esophageal carcinogen.

“Our data provide strong evidence that anthocyanins are important for cancer prevention,” said the study’’s lead author, Gary D. Stoner, Ph.D., a professor in the department of internal medicine at Ohio State University.

During the study, the team led by Stoner fed rats an anthocyanin-rich extract of black raspberries and found that the extract was nearly as effective in preventing esophageal cancer in rats as whole black raspberries.

The researchers conducted clinical trials using whole berry powder, which has yielded some promising results, but required patients to take up to 60 grams of powder a day.

“Now that we know the anthocyanins in berries are almost as active as whole berries themselves, we hope to be able to prevent cancer in humans using a standardized mixture of anthocyanins,” said Stoner.

“The goal is to potentially replace whole berry powder with its active components and then figure out better ways to deliver these components to tissues, to increase their uptake and effectiveness.

“Ultimately, we hope to test the anthocyanins for effectiveness in multiple organ sites in humans,” he added.

FDA questions Lilly, Amgen cancer drug trials

U.S. health regulators are questioning whether clinical trials proposed by ImClone Systems and Amgen Inc will be adequate to show that patients with a certain type of gene are more likely to be helped by two cancer drugs, according to documents released on Friday.

Early research has indicated that patients with the normal, so-called “wild type” of a gene known as K-ras respond far better to treatment with ImClone’s Erbitux and Amgen’s Vectibix than those with a mutated form of the gene.

On Tuesday, the U.S. Food and Drug Administration will seek advice from outside experts on what type of clinical trial the companies need to conduct to confirm the findings.

While eliminating patients with a mutated K-ras gene from treatment would cut sales of the drugs, the findings could be a significant advance toward so-called specialized medicine in which genetic testing can determine who is most likely to benefit from expensive drugs.

ImClone, recently acquired by Eli Lilly and Co. and Bristol-Myers Squibb Co, co-market Erbitux in the United States.

Both ImClone and Amgen have proposed retrospective testing of K-ras status from large randomized trials that have already been designed and have recruited patients or are still looking to enroll them, the FDA said in documents released ahead of Tuesday’s meeting.

The drugmakers modified the ongoing studies to enroll only patients with the wild type K-ras gene, the FDA said.

Amgen has already enrolled patients in two, large ongoing studies but made changes before conducting any K-ras testing, the company said in a separate document. Results are expected next year.

Enrollment in two earlier trials for ImClone’s Erbitux were halted after the K-ras finding so patients could be informed. Patients are still being sought for another ongoing trial that was put on hold, the company said.

“It is no longer feasible to conduct clinical trials in patients with MT (mutant) K-ras in their tumors,” it wrote.

Amgen shares were up 0.6 percent at $57.59 on Nasdaq. Lilly shares were also up 0.6 percent, at $35.84, and Bristol was up 3.5 percent at $22.21 on the New York Stock Exchange.
